In this study, the isolation, identification and antibiotic susceptibility of bacterial agents taken from urine and sediment samples of 10 cats and 10 dogs, which were diagnosed with urinary tract infection was aimed. Each urine and sediment samples were streaked onto blood agar which contains 5 % sheep blood, Eosin Methylene Blue agar and MacConkey agar for bacterial examination. In conclusion, Staphylococcus aureus from 4 dogs, Staphylococcus epidermidis from 2 dogs, Serratia liquefaciens from 1 dog, Plesiomonas shigelloides from 1 dog, Yersinia enterocolitica from 1 cat, were isolated and identified. In conclusion of antibiogram test, 78 % of the isolates were susceptible to Amoxicillin-Clavulanic acid, 67 % of the isolates were susceptible to Ceftriaxone and Cefoperazone, 55.5 % of the isolates were susceptible to Enrofloxacin, 22.5 % of the isolates were susceptible to Lincomycin and 22 % of the isolates were susceptible to Erythromycin. The isolates were resistant to Sulfamethoxazole-Trimethoprime and Oxacillin in the ratio of 78 % and 67 % respectively. Consequently, it is concluded that the isolates obtained from dogs highlight the resistancy to antibiotics, bacterial agents play a less role in feline urinary tract infections and the appropriate antibiotic application procedure will enhance the success of therapy.

Identification of bacterial pathogens that cause urinary tract&nbsp;infections and detection of their antibiotic susceptibility

<div>Major constraints in increasing lentil growing area in eastern India is mortality due to several diseases throughout the growth stages. Breeding lentil for disease tolerance by genotypic resistance is most promising and easier method. Therefore, a collection of 500 genotypes have been screened in this study for pre-flowering blight-like disease which is predominant in experimental field for last ten years. Fungal DNA isolation from ten randomly selected diseased plants followed by ITS sequencing using universal primer pair revealed cent percent similarity in NCBI database with single organism Alternaria alternata. Three popular varieties viz. WBL-77, Ranjan and Asha were kept as check for disease reaction comparison. 466 genotypes survived till flowering and scoring was made using 1-9 scale. Only three lines, EC866132, IC267667, IC201778 established as the tolerant lines against Alternaria blight attack at the pre-flowering stage.</div>

<div>Screening lentil (Lens culinaris Medik) Genotypes for Resistance against Pre-Flowering Blight and Identification of Pathogen by ITS Sequencing</div>

Crop pathogens pose a significant threat to global food security, causing substantial economic losses and food safety concerns. The identification and quantification of compounds produced by these pathogens are crucial for understanding their diagnosis by biochemistry, virulence mechanisms and predict for disease control. Initially, diagnosed by mass spectrometry such as GCMS and LCMS are powerful analytical techniques used for the identification and quantification of compounds in various fields, including environmental science, pharmaceuticals and food analysis. This study aims to explore the application of mass spectrometry for the diagnosis of crop pathogens, focusing on the identification and quantification of volatile organic compounds (VOCs) and secondary metabolites produced by fungal and bacterial pathogens. The study will also evaluate the advantages and disadvantages of GCMS and LCMS for crop pathogen analysis, including sensitivity, resolution, selectivity, range of applications, sample preparation requirements, cost and data interpretation challenges. The results will provide valuable insights into the potential of GCMS and LCMS for crop pathogen diagnosis and highlight areas for further research and development in this field.

Mass Spectrometry in Crop Pathogens Diagnosis: A Comprehensive Review

<div style="text-align: justify;">Environment is an important aspect of plant ecology and global environmental change is of major concern that is caused by natural and human activities which alter greenhouse gas concentrations in the atmosphere. The increase in concentration of greenhouse gases is foreseen to continue to raise average global temperature. Elevated concentration of carbon dioxide with increased temperature influences the plant-disease interactions. The environment has an influence over the development as well as temporal and spatial dissemination of plant diseases. The result of change in environment can either be favorable, non favorable or impartial, as these changes can either lessen, expand or have no influence on diseases as each disease may be attributed differently to these variations in accordance to an area or time of year. Variation in environmental conditions is said to be influencing plants in natural ecosystems all around the world and change in climate directly impacts crops, along with their synergy in accordance with the microbial population. The important elements governing magnification and spread of plant diseases are temperature, moisture, light and carbon dioxide concentration. Environment change causes a significant impact on germination, reproduction, sporulation, spore dispersal, along with perforation by pathogens as a vulnerable host will not be invaded by a pernicious pathogen if the environmental conditions are not facilitative for disease. The environment influences all life stages of host as well as that of a pathogen and as a result induces an opposition to various pathosystems. Resistance mechanisms of plants, including effector-triggered immunity (ETI), pattern-triggered immunity (PTI) and defence network of hormones, are particularly influenced by environmental elements. Pathogenic virulence mechanisms like fabrication of virulence proteins and toxins, and also spore germination and survival are governed by factors such as atmospheric carbon dioxide, temperature and humidity. A large number of in-vitro experiments to understand interactions between plants and pathogens rely on predetermined pathosystems by making use of ascertained environmental conditions which take into consideration relatively small part of vital plant&ndash;pathogen&ndash;environment interactions occurring in natural ecosystem. Thus, there is a need to research effectiveness of disease management strategies that is, to assess current management strategies to understand multifaceted nature of environment-pathogen interactions for production of crops that are irrepressible to environment change.</div>

Environment-Pathogen Interaction in Plant Diseases

<div>Background: Illegal migration of pigs/piglets from Myanmar to Mizoram is a common practice to meet the local demands. The migrated animals are suspected as potential carrier of various microbial pathogens. The present study was conducted on isolation, identification and molecular characterization of major bacterial pathogens (Actinobacillus pleuropneumoniae, Bordetella bronchiseptica, Haemophilus parasuis, Mycoplasma hyopneumoniae and Pasteurella multocida) in pigs illegally migrated from Myanmar to Mizoram.&nbsp; 
 
Methods: A total of 209 rectal swabs and 209 nasal swabs were collected from apparently healthy migrated pigs during October 2018 to April, 2019. All the samples were processed for PCR based detection of target bacterial species followed by isolation and identification by bacteriological techniques. The bacterial species were further confirmed by BD Phoenix automated bacterial identification system and selected virulence genes of the bacterial species were determined by specific PCR assay.&nbsp; 
 
Result: By species specific PCR, 110 samples were found to be positive for selected bacterial species, of which 20 (9.57%), 1 (0.478%), 86 (41.15%), 2 (0.956%) and 1 (0.478%) were A. pleuropneumoniae, B. bronchiseptica, H. parasuis, M. hyopneumoniae and P. multocida, respectively. A total of 52 bacterial strains were isolated and identified, of which 11, 1, 39 and 1 were A. pleuropneumoniae, B. bronchiseptica, H. parasuis, M. hyopneumoniae and P. multocida, respectively. Virulence genes were detected in A. pleuropneumoniae and H. parasuis isolates. Based upon the published literatures, this is the first ever report of isolation and identification of pathogenic A. pleuropneumoniae and H. parasuis in pigs in India.</div>

Transmission of Novel Bacterial Pathogens through Pigs Transported from Myanmar to Mizoram

<div style="text-align: justify;">The aim of the present study is to identification and classification of different associated fungi from the seeds of Eruca sativa. Total 10 species of fungal pathogens belonging to 7 genera are isolated from the Saudi (Ss) and Indian seeds of Eruca sativa. Saudi seeds affected by the fungal pathogens namely, A. niger, A. flavus, A fumigates, Mucor spp., Pencillium spp., while Indian seeds affected by Aspergillus niger, A. flavus, A. fumigates, Aspergillus terreus, Pencillium spp., Mucor spp., Rhizopus stolonifer, Humicola insolens, Colletotrichum gloeosporioides, Fusarium oxysporum. However, many of the soil borne or seed borne associated fungal pathogens spread the diseases in plants as well as in human beings. So the present study reveals that, the proper identification of fungal pathogens and their symptoms are easily recognized. Then the prevention care could be taken during the agriculture and high yield produced before the spread of symptoms and diseases. &nbsp;</div>

<div style="text-align: justify;">Study of Associated Fungal Pathogens on Seeds of Eruca sativa Mill., (Gargeer)&nbsp;</div>

<div>Background: Pigeonpea [Cajanus cajan (L.) Millsp.], an important grain legume crop that is predominantly cultivated in tropical and sub-tropical regions of the world (Nene and Sheila, 1990). &nbsp;Crop wild relatives (CWR) are valuable source of novel alleles for economic traits which when transferred to cultivated species adapt well to changing biotic stresses, farming practices, market demands and climatic conditions. Under proposed study the detailed symptomatology, cultural, morphological and pathogenic characteristics of the causal agent and molecular identification of the pathogen were identified. 
 
Methods: Of the total 79 accession of wild relatives of pigeonpea (Cjanus cajan) planted in the experimental fields of ICAR- Indian Institute of Pulses Research (IIPR), Kanpur during 2017-18, one accession (ICP-817) of Rhynchosia bracteata was found to be affected by a disease characterized by leaf and stem blight symptoms. Disease symptoms appeared in January&ndash;February, 2017 and 2018. Presence of water soaked lesions on the leaves and stem which later turned chalky in appearance, cottony white mycelium on the affected plant parts along with black coloured irregularly circular sclerotia resembled those of white mold fungus Sclerotinia sclerotiorum (Lib) de Bary were &nbsp;observed. 
 
Result: Pathogenicity of the fungus was proved using detached leaf/twig inoculation techniques. Morphological characters and the sequences of internal transcribed spacer (ITS) region of the fungus confirmed the causal agent of the disease to be Sclerotinia sclerotiorum (Lib) de Bary. This is the first report of the Sclerotinia sclerotiorum on Rhynchosia bracteata not only in India but also in the world. &nbsp;</div>

<div>Identification and Characterization of Sclerotinia sclerotiorum (Lib.) de Bary Associated with Rhynchosia bracteata Benth. Ex Bak. of Wild Derivatives of Pigeonpea from India</div>

<div>Background: Fusarium wilt in lentil (Lens culinaris L.) infected by Fusarium oxysporum is considered as one of the major biotic factors for low productivity in lentil. The present study was conducted to determine nine Fusarium isolates (W1-W8 and W10) which were isolated from different lentil growing fields of West Bengal, India and were characterized for their physio-morphological, biochemical and molecular traits in vitro.&nbsp; 
 
Methods: Nine Fusarium infected lentil plant samples were collected from 9 different places of West Bengal. Pathogen was isolated from lentil samples using PDA media followed by cultural, morphological, pathogenic and molecular variability studies for all the isolates of Fusarium. Type of pigmentation, sporulation, conidia size of each isolate was recorded by observing culture plate after complete growth of the mycelium. Pathogenicity test were done to check the aggressiveness of all the 9 isolates of Fusarium in three varieties of lentil (Maitri, HUL-57 and BM-6). The isolated fungal DNA was amplified with specific primer pair, Forward primer ITS 1 and Reverse primer ITS 4. 
 
Result: Morphological assessment of 9 isolates showed distinct identification characters and variability. The mycelia of all the isolates were septate, profusely branched with differential pigmentation, texture and margin.All the Fusarium isolates produced micro-and macro-conidia in pure culture within seven days after inoculation and showed different morphological characters in pathogenicity test, variety Maitri showed better germination (100% in W2, W4, W6, W7, W8 and W10) and less disease severity against most of the strains (75% in W1) as compared to other two varieties against and hence variety Maitri taken into consideration for greenhouse and field trials. Furthermore, identification of molecular variability was performed by sequence analysis of rDNA-ITS region of each isolate and sequencing of rDNA-ITS region supported the morphological study and confirmed the associated fungi at species level.&nbsp;</div>

Morpho-molecular and Pathogenic Variability of Wilt of Lentil from Indo-Gangetic Plains of India

<div style="text-align:justify">Background:&nbsp;Fusarium oxysporum f.sp. ciceris (Foc) is one of the most important pathogen, causing wilt of chickpea. It is soil and seed borne pathogen. &nbsp;A serious threat in Foc is the evolution of new races, which reduces exploitation of wilt resistance in the crop in a particular area. Eight races (race 0, 1A, 1B/C, 2, 3, 4, 5 and 6) were reported &nbsp;in the world and &nbsp;among them four races &nbsp;(race 1, 2, 3, 4) &nbsp;were &nbsp;from India. Race 1 was reported from Andhra Pradesh. It is very important to monitor the variation regularly in new isolates collected from different varieties or genotypes and different geographical regions to identify the racial pattern. Development of SCAR markers for identification of Foc isolates is also important as they are highly reliable.&nbsp; 
 
Methods: Twenty isolates of Fusarium oxysporum f.sp. ciceris were obtained &nbsp;from wilt infected plants of chickpea covering different places of Andhra Pradesh including two isolates from Telangana and confirmed the pathogen based on pathogenicity test. They were &nbsp;tested on &nbsp;host differentials of chickpea for &nbsp;races identification. Among these twenty isolates five most virulent isolates (Foc-6, Foc-10, Foc-12, Foc-17 and Foc-24) were selected for RAPD based on pathogenicity test and SCAR marker was developed based on DNA banding pattern during RAPD for one isolate. 
 
Result: Based on the disease reaction on differentials, concluded that 17 of them out of 20 were matched with race-1 reaction. Two isolates were &nbsp;matched with race-6 reaction and one is not matched with none of the races. An amplified product of polymorphic band of approximately 700 bp in the isolate &nbsp;Foc-12, obtained during RAPD analysis &nbsp;was selected for &nbsp;SCAR marker development and two SCAR markers were developed and validated. Identification of races mainly helps in development of resistant cultivars to specific races and might be contributed to development of integrated disease management practices for Fusarium wilt.</div>

<div>Identification of Races of Fusarium oxysporum f.sp. ciceris, Inciting Wilt of Chickpea in Andhra Pradesh and Parts of Telangana</div>

<div>Background: Mastitis, the inflammation of parenchyma of mammary gland is frequently considered to be costliest and complex disease prevalent in India. Mastitis is caused by pathogens like Staphylococcus spp., Streptococcus spp., Mycoplasma bovis, E. coli, Klebsiella spp., Citrobacter spp., Enterobacter spp. and Entercoccus. The treatment of mastitis in animals is carried out using antibiotics. Treatment failure in mastitis is due to increased antibiotic resistance of mastitis pathogens and also due to indiscriminate use of antibiotics without testing in vitro antibiotic sensitivity test against causal organisms. In comparison to cultural method, PCR assays takes less time for detection of bacteria from the mastitis milk samples. Present research work was carried out regarding isolation, identification and multiple drug resistance profile of clinical bovine mastitis associated pathogens using conventional as well as molecular approach.&nbsp; 
 
Methods: In the present study, 73 mastitis milk samples were collected from Anand and Panchmahal district of Gujarat. The milk samples were subjected for cultural isolation and DNA extraction for identification of bacteria by cultural and PCR method. Antimicrobial sensitivity pattern of the isolates were carried by disc diffusion method and isolates were categorized in multiple drug resistant. &nbsp; &nbsp;&nbsp; 
 
Result: In the present study, Out of 73 mastitis milk samples collected from cows 48 (65.75%) cows were positive for bacterial isolation and S. aureus was the most predominant bacterial species. PCR from the mastitis milk additionally detected bacteria in culturally negative milk samples. Most sensitive drug was gentamicin and most of the isolates (90.19%) showed the multiple drug resistance for the two to nine drugs with 0.1 to 0.6 multiple antibiotic resistance index.</div>
