CALLUS INDUCTION AND PLANT REGENERATION IN SORGHUM (SORGUM BICOLOR L. MOENCH)

The in vitro response of 15 sorghum genotypes were assessed on MS media containing 2, 4-D 2 mgl-1 and Kinetin 0.5mgl-1 (M1) for callus induction from immature inflorescence and shoot apex. Callus induction was observed only in six genotypes viz., SOR 499, SOR 530, SOR 668, SOR 984, AS 210 and AS 1619 from immature inflorescence and in five genotypes viz., SOR 499, SOR 530, MS 8388, MS 8445 and AS 1619 from shoot apex. These genotypes were tested on MS + 2, 4-D 3 mgl-1, Kinetin 1 mg-1 (M2), MS + 2, 4-D 2 mgl-1, Kinetin 1 mg-1 (M3) and MS + 2, 4-D 3 mgl-1, Kinetin 0.5 mg-1 (M4). Shoot apex did not show any proliferating callus on M3 and M4. The genotypes, SOR 499 and AS 1619 had the lowest callus induction days, highest per cent of callus induction and callus weight in MS media containing 2, 4-D 2 mgl-1+ Kinetin 0.5 mg-1. Plant regeneration was achieved on MS + IAA 5 mgl-1 + Kinetin 0.2 mgl-1. The highest percentage of plant regeneration was observed in genotypes SOR 499 and AS 1619.
