Background: The porcine sapelovirus (PSV) is a small, non-enveloped, single-stranded, positive-sense, RNA virus of the family Picornaviridae. The PSV infections in pigs have been found associated with diarrhoea, polioencephalomyelitis, pneumonia and reproductive disorders with a high morbidity rate. Despite of its economical importance very few studies are available on the pathology of PSV. The present study was conducted with the aim to investigate the PSV infection and associated pathology in Indian pigs.&nbsp; 
 
Methods: Tissue samples along with intestinal content were collected from a total of 78 necropsied cases for histopathological examination and molecular investigation during April 2019 to August 2020. The amplification of 5&#39; UTR region of PSV was carried out via RT-PCR and confirmed by sequencing. The Genetic characterization of Indian isolate of the PSV was done on the basis of viral 5&#39; UTR gene.&nbsp; 
 
Result: A total of eight out of 78 cases were found positive for the PSV. Catarrhal and haemorrhagic enteritis, thickening and clouding of brain meninges along with congestion of brain and pneumonia was observed as common gross lesions. Microscopic lesions included perivascular cuffing, focal gliosis, neuronophagia, congestion of meningeal and cerebral vessels, interstitial pneumonia, inflammatory changes in the intestinal mucosa and sloughing of villi. The genetic characterization revealed maximum identity of 96.89% with PSV-1 strain PSV-46-V (LC508233) and PSV-1 strain PSV-26-B (LC508232) of Zambia. This study reported the pathological and molecular investigation of PSV from Indian pigs. Further explorative surveillance along with experimental studies in suitable animal model and cell lines are highly warranted for better understanding of PSV pathology in Indian pigs.

Pathology and Molecular Characterization of Porcine Sapelovirus in Indian Pigs

<div>Background: Trichomonas gallinae is a flagellated protozoa, infects the upper digestive system of pigeon and causing a condition called canker, characterized by development of granulomatous lesions in the buccal cavity, crop, proventriculus, gizzard and liver of affected birds causing mortality upto 90% in squabs. Systemic research on prevalence and pathology of T. gallinae in pigeons are scant in India. The present work was carried out to document the prevalence and pathology of T. gallinae in pigeons of Assam. 
 
Methods: During the one- year study period, throat swab samples were collected from households, market places and temple premises in 4 districts of Assam. A total of 324 birds were examined by wet mount and Giemsa staining methods for detection of T. &nbsp;gallinae. Naturally died carcasses were studied for gross and histopathological alteration in different organs. 
 
Result:&nbsp;In the present study prevalence of T. gallinae was found 26.85% and squabs were highly susceptible with 56.25 percent. &nbsp;Sex wise, comparatively higher prevalence was observed in females (33.54%) than in the males (20.48%). Seasonality study revealed highest prevalence during winter. Gross pathological alteration characterized by yellowish white caseo-necrotic haemorrhagic lesion in the upper digestive tract beginning with the oral cavity and lesions also found in liver and lungs. Histopathological alteration also recorded in different organs. In the liver sinusoidal congestion and kupffer cell hyperplasia were observed. Infiltration of polymorphonuclear cells in the thickened inter alveolar space of lungs also recorded. Further study is required to identify the prevalent strains of T. gallinae on the basis of pathogenicity in this geographic region and immunology of T. gallinae in pigeons and other hosts remain to be explored.</div>

<div>Prevalence and Pathology of Trichomonas gallinae in Domestic Pigeon (Columba livia domestica) of Assam, India</div>

Background: A total of 20 tissue samples from horn, eye, skin and vulva in bovines suspected of squamous cell carcinoma (SCC) on the basis of gross pathology were collected from Durg, Dhamtari and Rajnandgaon districts of Chhattisgarh and processed in the Department of Veterinary Pathology, College of Veterinary Science and A.H., DSVCKV, Anjora, Durg (C.G.). 
&nbsp; 
Methods: Investigation was conducted during January to June 2022, we explicate the pathological and immunohistochemical examination including scoring and grading of squamous cell carcinoma in bovines. Histopathological examination as per standard HandE method of staining and immunohistochemistry was performed through Benchmark automated staining system.

Result: Twelve cases were diagnosed as SCC on the basis of histopathological and eleven on the basis of immunohistochemical analysis. Grossly neoplasms of horn revealed large, cauliflower like unilateral irregular masses at the base of horn and they were friable and soft in consistency. Small, unilateral growths observed in eye varied from soft to hard in consistency. Histopathological findings in SCC of horn revealed keratinization of horn epithelium forming concentric keratin pearls. Distinctive keratin pearls with more keratin deposition towards the center, numerous mitotic figures with anaplasia and neovascularization were evident in OSCC. Immunohistochemistry revealed strong immunohistochemical expression of Pan-Cytokeratin (Pan-CK), p53 and EGFR and negative&nbsp; to p16. Immunohistochemical scoring (IHS) was done in Pan-CK, EGFR and p53, ranged from 1-4. Well differentiated SCC of horn revealed strong immunoexpression against EGFR, varied from mild to intense. SCC of horn exhibited strong immunoexpression for Pan-CK, p53 and EGFR whereas, immunoexpression in OSCC specifically p53 was less as compared to SCC of horn. Grading and differentiation of SCC (horn and eye) was also done on basis of histopathological and immunohistochemical scoring.

Pathology and Diagnosis of Squamous Cell Carcinoma in Bovines

<div>Background: Paratyphoid infection of poultry is caused by non-host adapted motile salmonellae and are responsible for numerous cases of food borne illness worldwide. The present study was carried out from July 2019 to February 2020 in Jabalpur to know the occurrence and pathology of paratyphoid bacteria in poultry. 
&nbsp; 
Methods: Whole blood agglutination test was performed to know the prevalence of salmonellosis in and areas surrounding Jabalpur region and pooled fecal samples were collected from poultry farms to perform microbe culture and biochemical characterization. Serotyping of Salmonella isolates was done using polyvalent antisera. Necropsy examination was conducted to observe gross and histopathological lesions.&nbsp; 
 
Conclusion: Rapid whole blood agglutination test determined the percent prevalence of Salmonella as 28.0% from 25 private poultry outlets. The percent prevalence of salmonellosis by collecting pooled fecal samples from 15 broiler and 11 layer farms was recorded as 20.0% and 45.4% respectively. Salmonellosis was recorded in 1.58% of total necropsy cases of birds examined for gross and histopathological studies. Polyvalent antisera diagnosed 27.27% motile paratyphoid salmonellae, out of which 18.18% tested positive for Salmonella Enteritidis while 9.09% tested positive for Salmonella Typhimurium. Birds with paratyphoid infection showed hepatomegaly, discoloration, hemorrhagic and necrotic foci in liver and various grades of hemorrhagic to catarrhal enteritis were recorded.</div>

​Incidence and Pathology of Paratyphoid Infection in Poultry

<div>Background: This study elucidated the molecular detection and pathological alterations in broiler chickens naturally infected with field circulating NDV strains along with their phylogenomic dynamics. 
 
Methods: Morbid tissue samples of diseased/dead chickens were collected from 100 poultry flocks presented to poultry disease diagnostic laboratories from September 2018 to August 2019. Samples were subjected to molecular detection of NDV along with phylogenetic analysis and subsequent gross and histopathological examination. 
 
Result: Based on RT-PCR results, the positivity of NDV was 04/100 (4%). Genetic analysis of the NDV Fusion (F) gene revealed 98.92% and 98.74% similarity with Iranian and Pakistani isolates, respectively. The evolutionary tree showed that present study isolates were placed in a clade belongs to genotype Vll sub-genotype i and l. Necropsy examinations revealed the petechial haemorrhages associated with multifocal necrosis in gastrointestinal and respiratory organs. Besides these pathological findings, amino acid sequence of F gene revealed that study isolates are having pathogenic potential similar to the velogenic strains of NDV. Based on all essential analyses, the present study concluded that the evolution and distribution of the Newcastle disease virus of various genotypes VIIi and VIIl in Pakistan are having significant pathogenic potential. Therefore, it emphasizes developing ND vaccine from indigenous strains for better protection of commercial poultry in Pakistan.</div>

Molecular Characterization and Pathology of Virulent Newcastle Disease Virus Isolated from Broiler Chickens in District Lahore, Pakistan

<div>Background: Postweaning multisystemic wasting syndrome (PMWS) is caused by porcine circovirus type 2 (PCV2). It is a globally emerging clinical condition which affects nursing piglets and growing pigs. PCV2 requires various cofactors for fully development of PMWS. The present report describes the pathology of PMWS associated with classical swine fever virus (CSFV), porcine parvo virus (PPV) and multidrug resistant enteric Escherichia coli infections in an organized swine farm, Rajasthan. In India, this is the first report on pathology of PMWS associated with all the three viruses and multidrug resistant enteric colibacillosis. 
 
Methods: Detailed necropsy was conducted on pig carcass submitted to Post-mortem Facility, Division of Pathology, ICAR-Indian Veterinary Research Institute (IVRI), Izatnagar in the year 2017 from an organized swine farm, Rajasthan, India. Heart blood and diarrhoeic intestinal contents were collected aseptically and stored separately at 4&deg;C for bacteriological examination and antibiotic sensitivity test. Tissue samples were collected in sterile container and stored at -40&deg;C for DNA and RNA extraction and polymerase chain reaction (PCR) and also collected in 10% neutral buffered saline for histopathological processing. 
 
Result: The organized farm was having history of 33% (200/600) of mortality among 2 months to 6 months old piglets. Gross lesions were wasting, lymphadenopathy, non-collapsed lungs, hydropericardium, hepatomegaly, icterus, enteritis and pale kidneys, button shaped ulcers in the caecum and colon etc. Histopathologically, lymphoid depletion, reticuloendothelial cell proliferation in lymphoid tissue, fibrin thrombi in spleen, severe hepatitis, interstitial pneumonia, interstitial nephritis, severe necrosis in large intestine, meningitis, perivascular cuffing (PVC), gliosis, neuronal degeneration in cerebrum etc were observed. Molecular diagnosis and bacteriological examination confirmed presence of PCV2, PPV, CSFV and multidrug resistant Escherichia coli.</div>

Pathology of PMWS Associated with Classical Swine Fever Virus, Porcine Parvo Virus and Multidrug Resistant Enteric Escherichia coli Infections in an Organized Swine Farm, Rajasthan, India

<div>Background: Pyometra is a pathological state of the compromised uterus of grown-up to old, diestrum bitches. Pyometra is easy to identify but can be more challenging when there is no vaginal discharge. In the present study, an attempt was made to find out the pathogen involvement and clinico-haemato-biochemical alteration in bitches affected with Pyometra. \ 
 
Methods: Twelve bitches with a history of inappetence, polydipsia, dirty vaginal discharge, occasional vomition irrespective of breeds and age groups were clinically examined and diagnosed as pyometra. The presumptive clinical diagnosis was based on case history, clinical signs, vaginal cytology, abdominal ultrasonography or radiography, gross examination of a pus-filled uterus after the ovariohysterectomy. 
 
Result: Vaginal examination revealed brown chocolate malodorous vaginal discharge with discolouration of the perivulval tissue. Abdominal ultrasound showed hyperplasia with echogenic material in the uterine wall along with distended uterine horns containing a large volume of cellular contents. Radiographic imaging revealed cranio-dorsal displacement of the small intestine and a twisted uniform tubular opacity with a distended uterus. The vaginal cytology revealed a large number of degenerated neutrophils, intermediate and parabasal cells. Escherichia coli were the major pathogen isolated from the pyometra uteri of bitches. The surface of the endometrium was columnar ciliated epithelium, with hyperplasia of subepithelial tissue and thick muscularis. The haemato-biochemical evaluation in pyometra infected bitches revealed decreased haemoglobin (Hb), packed cell volume (PCV), total erythrocytic count (TEC) and lymphocyte count indicating the microcytic hypochromic type of anaemia. Leucocytosis with a predominant absolute neutrophilia and lymphocytosis were consistently found in canine pyometra. There was a significantly elevated profile of blood urea nitrogen, creatinine, transaminases and alkaline phosphatase as well as total proteins observed in pyometric female dogs as compared to healthy ones which may have diagnostic and prognostic importance, which also indicated damage of vital organ caused by bacterial endotoxins. The haemato-biochemical indices correlating with imaging techniques, vaginal cytology and histopathological examination can serve as good prognostic markers in bitches with pyometra.</div>

Clinico-haemato-biochemical and Pathological Alteration of Pyometra in Canines

Background: Fatty Liver Hemorrhagic Syndrome (FLHS) is an important non-infectious metabolic syndrome in laying hens, with excessive fat retention in the liver, grossly liver rupture and sudden death. 
 
Methods: This study examined the prevalence, pathology and risks of FLHS in Kadaknath chickens raised on various management systems within the Rewa district of the Madhya Pradesh between 2022 and 2024. There were 830 birds that were studied, both backyard and organized farm flocks. To identify and describe lesions, necropsy and histopathological analyses were conducted to establish diagnosis. 
 
Result: The total prevalence of FLHS was 12.65%, with early laying birds (21-30 weeks) showing the highest incidence (18.2) at the period when estrogenic activity was high and the hepatic lipogenesis was at its peak. The seasonal patterns also showed that it prevailed more in the winter (19.2%), then summer (10.6%) and the rainy seasons (9.3%). Gross pathological alterations were enlarged yellowish friable livers with subcapsular and intraparenchymal hematoma, excess abdominal fat and ovarian degeneration. Microscopically the affected liver showed severe lesions of vacuolar degeneration of hepatocytes, sinosoidal congestion and multifocal hemorrhages which confirmed hepatic steatosis with vascular compromise. The results indicate that predisposing factors are early stages of lay, high energy diets, low mobility and metabolic stresses associated with seasons. Although Kadaknath chickens are famous in terms of resisting, the prevalence observed did not imply that such chickens are resistant to FLHS in intensive or semi-intensive farms. The dietary lipid modulators, better management and environmental control should be implemented to reduce the risk. The present study supplies baseline epidemiological and pathological data concerning FLHS in Kadaknath, which contributes to the establishment of breed-specific preventive actions aimed at increasing the productivity and welfare.

Prevalence and Pathology of Fatty Liver Hemorrhagic Syndrome in Kadaknath under Different Rearing Systems

<div>Background: The thyroid gland is vital endocrine gland which secretes three hormones i.e. thyroxin (T4), triiodothyronine (T3) and Calcitonin hormones. The thyroxin (T4) and triiodothyronine (T3) hormones are biologically active and are required for maintenance of normal levels of metabolic activity. The thyroid also produces calcitonin from the parafollicular cells which act directly on osteoclast to decrease the bone resorption which lower the blood calcium level. Deficient or excessive production of thyroid hormones may lead to serious pathological states with outward symptoms.&nbsp; 
 
Methods: The gross anatomical and biometrical studies were conducted on thyroid gland of buffalo, sheep and goat (n=12) collected immediately after slaughtering from slaughter house and local meat shop. The weight of thyroid gland was measured by weighing balance, volume by water displacement method, length and width of lateral lobes and isthmus by calibrated scale and inelastic thread and thickness of lateral lobes and isthmus was measured by digital vernier calliper. The data was analysed statistically. 
 
Result: The lateral lobes were roughly triangular in buffalo and elongated in sheep and oval in goat. The surfaces were granular and rough in buffalo but smooth in sheep and goat. It extended from thyroid cartilage to 2nd tracheal ring in buffalo, 1st to 6th tracheal ring in sheep and 1st to 7th tracheal ring in goat. The left lobe was larger than the right lobe in all the three species studied. The thyroid gland was biggest in buffalo followed by goat and smallest in sheep. The density of isthmus was more than the lobes in sheep and goat but not in buffalo.</div>

​Comparative Gross Anatomical and Biometrical Studies on Thyroid Gland of Buffalo, Sheep and Goat

<div>Background: Bovine anaplasmosis is a haemolytic disease of cattle caused by an obligate intra-erythrocytic bacterium Anaplasma marginale and is characterized by high fever, dyspnoea, anaemia, icterus, decreased milk production and death. The present study was undertaken to study the gross and histopathological changes caused by naturally occurring A. marginale infection in cattle.&nbsp; 
 
Methods: History and clinical examination of animals revealed inappetance/anorexia, presence of tick infestation, high fever, pale or icteric mucous membranes, tachycardia, dyspnoea and lymphadenomegaly. The diagnosis of A. marginale infection was done by blood smear examination and further confirmed by polymerase chain reaction and sequencing analysis. During the present study, two animals did not respond to treatment and were succumbed to A. marginale infection. A systematic post-mortem examination was performed on the animals and gross lesions were recorded. The tissue samples from various vital organs such as heart, lungs, liver, spleen and kidneys were collected in 10% neutral buffered formalin and processed for histopathological examination.&nbsp; 
 
Result: The post-mortem examination revealed emaciated carcass with yellowish discoloration of serosal surfaces of the abdominal organs, haemorrhagic heart, congestion of lungs, hepatomegaly, distension of the gallbladder and splenomegaly. The major histopathological changes noticed included myocardial degeneration and necrosis, interstitial pneumonic changes, degenerative changes in hepatocytes and renal tubular epithelial cells, enlargement of red pulp area of spleen with histiocytic proliferation and hemosiderosis. Histopathology also revealed inflammatory process characterized by infiltration of mononuclear cells in various organs. Very few reports are available on the pathological aspects of A. marginale infection and thus the outcome of the present study can yield valuable information to aid in the diagnosis of bovine anaplasmosis under field conditions.</div>
