CLONING OF LEPTOSPIRA CAN/COLA SEROVAR GENE IN E. COU AND USING AS A PROBE IN SLOT BLOT HYBRIDIZATION
1ICAR Scheme on DNA based diagnostics for Leptospirosis.
Institute of Animal Health and Veterinary Biologicals, Hebbal, Bangalore . 560 024, India
ABSTRACT
Gene fragment of Leptospira serovar Canicola was cloned in E. coli cells using pUC18
plasmid as a vector. The recombinant cells were selected on LB agar medium, which contained
ampicillin, X-gal and IPTG. From the recombinant white colonies, plasmid was extracted and
the leptospiral DNA insert was collected after digestion with EcoR I and used as probe in
hybridization experiments.
